Liofilchem Kanamycin Aesculin Azide Agar Culture Media 620190 is a Liofilchem microbiology product supplied by Kormay Biomedicals. Application fit for the lab
This product fits environmental monitoring where repeatable sampling and neutralization are more important than a single isolated result. The lab should connect the medium to surface type, disinfectant residues, incubation plan, alert/action limits, and trend review.
Expected colonies or reaction pattern
Round white or grey colonies about 2 mm diameter surrounded by a black halo (aesculin reaction) are considered to be enterococci.
Principle behind the medium
Pancreatic digest of casein provides amino acids, nitrogen, carbon, vitamins and minerals for organisms growth. Yeast extract is a rich source of vitamins of B-group. Sodium chloride maintains the osmotic balance of the medium. Aesculin and ferric ammonium citrate are the indicator system. Sodium azide and kanamycin are the selective agents. Agar is the solidifying agent.
How to build it into the SOP
- Procedure: Inoculate the medium by spreading the sample over agar surface. Incubate aerobically at 35 ± 2°C for up to 3 days. An higher incubation temperature, i.e. 42 ± 0.5°C, may be used to increase selectivity.

- Quality control: Plates are inoculated with the microbial strains indicated in the QC table. Inoculum for productivity: 50-100 CFU. Inoculum for selectivity: 103-104 CFU. Incubation conditions: 18-24 h at 35 ± 2°C, in aerobic atmosphere. WARNING AND

- Warnings / precautions: The powder contains substances classified as hazardous under current legislation. The prepared medium is not classified as dangerous. In both cases, it is recommended to consult the safety data sheet before use. The product is intended for professional use only and must be used by properly trained operators.
- Storage / shelf life: The powder is very hygroscopic, store the powder at 10-30°C, in a dry environment, in its original container tightly closed. Store bottles at 2-8°C away from light. Do not use the product beyond its expiry date on the label or if product shows any evidence of contamination or any sign of deterioration.
- Waste disposal: of waste must be carried out according to national and local regulations in force.
