Result signals and confirmation
Clostridium perfringes is an anaerobic non motile organism, which generally reduces nitrate to nitrite (*). Motility testMotility is indicated by a diffuse growth extending out in the medium from the stabbing line.Nitrate testAdd a few drops of Alpha Naphtylamine and Sulphanilic Acid reagents (ref. 80257).Developing of a deep red colour indicates a positive test.NB. (*) Not all C. perfringens strains reduce nitrate to nitrite. Nitrate reduction occurs under anaerobic conditions only.
Method principle
Enzymatic digest of casein and meat extract supply amino acids, nitrogen, carbon, minerals, vitamins and other nutrients which support the growth of microorganism. Galactose is the fermentable carbohydrate. Potassium nitrate serves as base for the nitrate reduction test.Disodium phosphate acts as buffer. Agar is the solidifying agent included to make the medium semisolid and thus suitable for the determination of motility.Glycerol Supplement (ref. 80021) is added to the medium to improve the nitrate reduction reaction.
Bench workflow
- Preparation: Suspend 19.5 g of powder in 1 liter of deionized or distilled water and add 5 ml of Glycerol Supplement. Bring to boil and shake until completely dissolved. Distribute 10 ml per tube. Sterilize at 121°C for 15 minutes.NB. If the medium is not used within 8 hours from
- Procedure: Stab inoculate the medium with a pure culture of the organism being tested. For example, inoculate with isolated black colonies of C. perfringes from Tryptose Sulfite Cycloserine Agar (ref. 163872). Incubate anaerobically at 35 ± 2°C for 18-24 hours.
