Use case and sample types

Decision criteria after incubation
Lactose -fermenting organisms grow as pink to brick-red colonies , while lactose –nonfermenting organisms grow as colorless or clear colonies. Enterococci produce tiny red colonies. Although MAC CONKEY AGAR w/o BILE SALT is used for isolating and differentiating enteric microorganisms, biochemical and , if indicated, serological testing using pure cultures are recommended for complete identification.
Method logic
Peptones are sources of nitrogen and other nutrients. Lactose is fermentable carbohydrate. When lactose is fermented, a local pH drop around the colony causes a color change in the pH indicator (neutral red). Crystal violet is selective agents that inhibits growth of gram- positive organisms. Agar is the solidifying agent.
Preparation and execution notes
- Preparation: Suspend 50,0 g in 1 litre of distilled water . Heat until completely dissolved. Dispense into final containers. Autoclave at 121°C for 15 minutes.
- Procedure: Inoculate the specimen, in order to obtain isolated colonies, streaking it onto the suface of the medium using a sterile loop. Incubate at 36°C ± 1°C for 18-24 hours.
