When this format is useful
This product fits environmental monitoring where repeatable sampling and neutralization are more important than a single isolated result. The lab should connect the medium to surface type, disinfectant residues, incubation plan, alert/action limits, and trend review.
What to read after incubation
The formation of a yellow to brick red color indicates ammonia production. A negative reaction shows no color change. Other confirmatory tests, such as Oxidase Test and King’s B Medium may be required.
Selective or enrichment logic
Potassium phosphate is the buffering agent. Magnesium sulfate, sodium molybdate and iron sulfate are inorganic salts essential for organisms growth. Acetamide is the sole source of carbon in the medium. Sodium chloride maintains the osmotic balance of the medium. Acetamide is deaminated by gram-negative non- fermenting bacteria to produce ammonia.
Sample handling workflow
- Procedure: Filter volumes of diluted or undiluted water sample through 0.45 µm sterile membranes. Place each membrane on a plate of Pseudomonas CN agar and incubate at 36 ± 2°C for 40-48 hours. Count all colonies that produce blue/green (pyocyanin) color as confirmed P. aeruginosa. Acetamide Broth is used for confirming fluorescent non-pyocyanin producing colonies and reddish brown pigmented colonies: •Transfer all colonies to Nutrient agar and incubate at 36 ± 2°C for 20-24 h; •Inoculate a tube of Acetamide broth with the subculture and incubate at 36 ± 2°C for 20-24 h; •Add 1-2 drops of Nessler's Reagent and examine for production of ammonia.
