Method principle

Amies Transport Medium w/o Charcoal provides a reduced environment due to the presence of sodium thioglycollate and the small amount of agar. The Chloride salts supply essential electrolytes for transport and osmotic balance. Phosphates act as a buffer system. Sodium thioglycollate lowers the redox potential of the medium, thereby allowing a better conservation of anaerobic bacteria. The survival of bacteria in a transport medium depends on various factors such as bacteria type and concentration in the specimen, transport medium formulation, the temperature and duration of transport, and inoculation to appropriate culture media within 24 hours. Optimal growth and typical morphology can only be expected if direct inoculation and appropriate cultivation are followed.

Bench workflow

  • Preparation: Suspend 9.8 g of powder in 1 liter of distilled or deionized water. Heat to boiling, shake until completely dissolved and dispense into final containers as appropriate. Sterilize in autoclave at 121°C for 15 minutes.

  • Procedure: After collecting the clinical specimen with a swab, insert one third of the swab into the center of the medium. Then cut the stick and screw down the cap to force the swab to the bottom of the medium. Storage, limits, and safety

  • Limitations: • Some growth of contaminants may also occur during the long period of transport. • After transportation, the specimen should be inoculated in the proper medium as soon as possible. For optimum results, the time lapse between sample collection and inoculum onto culture medium should be reduced to the minimum. • It may not be suitable for the transport of fastidious organisms. • Gonococci survive well in Amies Transport Medium for at least 6 to 12 hours provided they are not exposed to temperature extremes.

  • Storage / shelf life: The powder is very hygroscopic: store the powder at 10-30°C, in a dry environment, tightly closed in its original container and use it before the expiry date on the label or until signs of deterioration or contamination are evident. Store prepared tubes at 2-8 °C.
