Method principle
Peptone, tryptone and meat extract provide nitrogen and other nutrient to support microbial growth. Yeast extract is a source of amino acids and vitamins of group B. Sodium chloride maintains the osmotic balance of the medium. Lecithin and sodium bisulfite aim to neutralize the preservatives commonly present in cosmetics.
Bench workflow
- Preparation: Suspend 37.8 g of powder in 1 liter of distilled or deionized water. Add 5 mL of Tween 80 (ref. 80031). Heat until completely dissolved. Dispense in final containers. Sterilize in autoclave at 121 °C for 15 minutes.
- Procedure: ! Inoculate 9 mL of LETHEEN BROTH BASE MODIFIED with 1 g or 1 mL of sample to test. Use larger volumes of broth if high levels of contamination are expected. ! Mix well to homogenize the suspension. ! Incubate at 32.5 ± 2.5 °C for at least 20 hours. ! Using a sterile pipette transfer 0.1 to 0.5 mL of suspension on a plate containing a nutritive medium and spread the sample onto the entire surface using a sterile loop. ! Incubate at 32.5 ± 2.5 °C for 48-72 hours.
Storage, limits, and safety
