Liofilchem E.M.B. Agar w Lactose + SUCROSE Culture Media 610140 is a Liofilchem microbiology product supplied by Kormay Biomedicals. Its practical role is selective medium for isolation of enteric Gram-negative bacteria from clinical specimens. What to read after incubation

At the end of the incubation period evaluate colonies characteristics as follows: Colonies of lactose fermenters are blue-black with or without a green metallic sheen. E. coli typically produces dark centered colonies with a green metallic sheen. Enterobacter spp. appear as brown to blue-black, mucoid colonies with no sheen. Colonies of non-lactose fermenting bacteria, such as species of Shigella and Salmonella, are colorless and translucent.

Selective or enrichment logic

The eosin dye inhibits growth of gram-positive bacteria and combines with the methylene blue indicator to produce a color change whenever lactose or sucrose are fermented. This medium does not allow discrimination between which carbohydrate is fermented. For example, Yersinia enterocolitica, which ferments sucrose, but not lactose, will produce the same purple-black colony as lactose-fermenting bacteria.

Sample handling workflow

  • Preparation: Dehydrated medium Suspend 38.0 g of the powder in 1 liter of distilled or deionized water. Mix well. Heat to boil shaking frequently until dissolved completely. Sterilize in autoclave at 121°C for 15 minutes. Cool at 45-50°C, mix well avoiding foam formation and aseptically distribute into Petri dishes.
