Application fit for the lab
Use this product where indicator organisms are part of a release, trend, or sanitation decision. The practical value comes from separating countable or presumptive target reactions from background flora so the analyst can document whether the sample meets the site limit.
Expected colonies or reaction pattern
Lactose-fermenting microorganisms produce pink to red colonies that are generally surrounded by a reddish zone of precipitated bile. Non-lactose-fermenting microorganisms result in colourless colonies. Beta-glucuronidase positive strains of E. coli exhibit blue fluorescent halos when examined under long-wave UV light.
Principle behind the medium
Peptone provides amino acids, nitrogen, carbon, minerals and vitamins for organisms growth. Lactose is a carbohydrate source. Yeast extract is a source of vitamin, particularly of B-group. Bile Salts Mixture and Crystal Violet inhibit Gram-positive bacteria. Neutral Red is the pH indicator. MUG (4-methylumbelliferyl-beta-D-glucuronide) is a substrate used for detecting glucuronidase activity. Agar is the solidifying agent.
How to build it into the SOP
- Preparation: Suspend 41.6 g of powder in 1 liter of deionized or distilled water. Bring to boil and shake until completely dissolved. Avoid overheating. DO NOT AUTOCLAVE. Cool up to 45-50°C. Aseptically, pour in Petri dishes.
- Procedure: Inoculate and incubate the plates at 35 ± 2°C for 22-26 hours.
