Decision criteria after incubation
The typical colonies of Campylobacter are small, greyish and flat with irregular edges. Colonies have a tendency to spread, especially when initially isolated from fresh clinical specimens.
Method logic
Peptospecial provides amino acids, nitrogen, carbon, minerals, vitamins and other nutrients which support the growth of microorganisms. Corn starch is a source of energy. Activated charcoal helps to neutralize the toxic metabolic products formed in the medium. Sodium chloride maintains the osmotic balance of the medium. Agar is the solidifying agent.Supplementation with Campylobacter Karmali Supplement (ref. 81036), containing cefoperazone, vancomycin, cycloheximide, hemin and sodium pyruvate, inhibits the contaminant microbial flora in fecal specimens improving, at the same time, the recovery of Campylobacter spp.
Preparation and execution notes
- Preparation: Suspend 48.0 g of powder in 1 liter of deionized or distilled water. Bring to boil and shake until completely dissolved. Sterilize at 121°C for 15 minutes. Cool up to 45-50°C. Aseptically, add rehydrated content of 2 vials (10 ml) of Campylobacter Karmali Supplement. Pour in Petri dishes.
- Procedure: Inoculate the plates by streaking directly the sample onto the agar surface. Incubate under microaerobic atmosphere (containing approximately 5% O2, 10% CO2 and 85% N2) at 42°C for a minimum of 48 hours.
