Buffered Yeast Extract (BYE) Broth is used for cultivating members of the Legionella genus, which require cysteine and iron to thrive. This medium is the charcoal-free broth version of Buffered Charcoal Yeast Extract (BCYE) agar, which is recommended by ISO 11731 for enumeration of Legionellae in water samples. TYPICAL FORMULA* (Per Litre of Purified Water) Yeast Extract 10.0 g α-Ketoglutarate 1.0 g ACES Buffer 10.0 g L-Cysteine HCl 0.4 g Iron(III) Pyrophosphate 0.25 g Final pH 6.9 ± 0.2 at 25°C *Adjusted and/or supplemented as required to meet performance specifications.
