Tryptone and peptone are sources of carbon, nitrogen, aminoacids and proteins. Glucose is the substrate for fermentation. Potassium phospahte is the buffer of the medium . Methyl red-positive organisms produce high levels of acid during fermentation of glucose, overcome the phosphate buffer system and produce a red color upon the addition of the methyl red pH indicator.
