Description
Skim Milk can be used to prepare Skim Milk Agar for detecting proteolytic microorganisms in foods, including dairy products. It can also be used to prepare Litmus Milk, a differential test medium for determining lactose-fermentation and for detecting proteolytic enzymes that hydrolyze casein and cause coagulation. This medium is particularly suitable for differentiating Clostridium species. When prepared in a 10% solution, it is equivalent to fresh skim milk and is used for the cultivation of lactobacillus.
Principle
Skim Milk is a source of lactose and casein in prepared culture media.
Preparation
- Mix the powder to a smooth paste with a small quantity of distilled water, then gradually add more distilled water until a 10% w/v mixture is obtained
- Sterilize at 121°C for 5 minutes
- The solution can be added to a culture medium or used by itself.
