Tryptone Water - Rev.0 / 07.08.2015. . Description. Tryptone Water is a liquid medium used for the differentiation of microorganisms based on indole production. This medium is recommended by ISO 7251 and ISO 3811 for the presumptive identification and enumeration of Escherichia coli in food and by the APHA for examination of water and wastewater.. Preparation. Dehydrated medium Suspend 15 g of the powder in 1 liter of distilled or deionized water. Heat to boil shaking frequently until completely dissolved. Sterilize in autoclave at 121°C for 15 minutes. TEST PROCEDURE Inoculate the tubes with pure cultures of the microorganisms to be tested.
