Method logic
Tryptone provides amino acids, nitrogen, carbon, minerals and vitamins for organisms growth. Dextrose is the carbohydrate source. Starch aids in neutralizing toxic metabolites. Bromocresol purple is the pH indicator. Agar is the solidifying agent.
Preparation and execution notes
- Preparation: Suspend 32.0 g of powder in 1 liter of deionized or distilled water. Bring to boil and shake until completely dissolved. Sterilize at 121°C for 15 minutes. Cool up to 45-50°C. Aseptically, pour in Petri dishes.
- Procedure: Inoculate and incubate the plates. The temperature and incubation period will vary according to the exact purpose of the investigation. For example, for enumeration of Mesophiles incubate at 32°C for 72 hours whereas for Thermophiles at 55°C for 48 hours.
