Bench workflow
- Procedure: With a glass spatula spread from 0.1 to 1.0 ml of the specimen or of diluted food (macerated in 0.1% Peptone Water) onto the surface of a well dried plate. Incubate at 36 ± 1 °C for 24-48 hours. Staphylococcus aureus is able to reduce tellurite to metallic tellurium resulting in growth as black colonies surrounded by a yellow zone.

- Formula / composition: (g/l) Tryptone 10.0 Yeast Extract 5.0 Mannitol 10.0 Dipotassium Phosphate 5.0 Lithium Chloride 5.0 Glycine 10.0 Phenol Red 0.025 Agar 15.0 Final pH = 7.2 ± 0.2 at 25 °C. DIRECTIONS Suspend 60.0 g of powder in 1 liter of distilled or deionized water. Heat until completely dissolved. Sterilize in autoclave at 121°C for 15 minutes. Cool to 45-50°C and aseptically add 20 ml of Potassium Tellurite 1% supplement (code 80022). Final medium will contain 0.01 mg of potassium tellurite per litre. Dispense in petri dishes.
