Result signals and confirmation
Take a 10 µl loop from turbid tubes, streak onto the surface of the Enterobacter (Cronobacter) sakazakii Isolation Agar plate (ref. 11623). Incubate the plates at 44°C for 18-24 hours. Examine for the presence of typical colonies of Cronobacter sakazakii, which are small to medium sized, green to blue-green coloured.
Method principle
Enzymatic Digest of Casein and Animal Tissue provides nitrogen, vitamins, minerals and amino acids essential for organisms growth. Lactose is the fermentable carbohydrate. Potassium phosphates are the buffering agents. Sodium chloride in addition to supply essential electrolytes for the osmotic balance, at a so high concentration restricts the growth of other members of the Enterobacteriaceae. Also sodium lauryl sulfate acts as selective agent inhibiting organisms other than coliforms. Vancomycin is added to the medium to inhibit Gram-positive bacteria.
Bench workflow
- Preparation: Suspend 64.6 g of powder in 1 liter of deionized or distilled water. Bring to boil and shake until completely dissolved. Sterilize at 121°C for 15 minutes. Cool to 45-50°C. Aseptically, add the rehydrated content of 2 vials (10 ml) of Vancomycin (5 mg) Supplement (ref. 81064) to get 10 mg/l vancomycin in the complete medium. Dispense into 10 ml tubes.
- Procedure: Homogenise the sample diluted 1:10 in Buffered Peptone Water (ref. 24199) and incubate at 37C for 16-20 hours. Transfer 0.1 ml of thebroth culture into a tube of Modified Lauryl Sulfate Tryptose Broth (mLST). Incubate at 44°C for 24 hours.
