Liofilchem Tryptic (CTA) Medium Culture Media 610185 is a Liofilchem microbiology product supplied by Kormay Biomedicals. Its practical role is tRYPTIC (CTA) MEDIUM is a semisolid medium used with added carbohydrates in differentiating microorganisms based on fermentation reactions and motility. Fermentation of the test carbohydrate is observed when acid is formed and the medium turns from red to yellow. Motility of an organism is evident as a haze of growth extending into the agar from the stab line.

How to build it into the SOP
- Procedure: Heavily inoculate the specimen by stabbing using a sterile needle and incubate at 36 ± 1°C for 18-24 hours.
Composition and QC checkpoints
- Formula / composition: (g/l) Tryptose 20.0 L-Cystine 0.5 Sodium Chloride 5.0 Sodium Sulphite 0.5 Phenol Red 0.017 Agar 2.5 Final pH = 7.3 ± 0.2 at 25 °C. DIRECTIONS Suspend 28.5 g of powder in 1 liter of distilled or deionized water. Heat to boiling until completely dissolved. Dispense into 10 ml tubes and sterilize in autoclave at 121 °C for 15 minutes. Cool to 45-50 °C. Aseptically add a sterile solution of carbohydrate at 0.5-1% final concentration. Mix well.
Handling limits and documentation
- Limitations: Neisseria species usually produce acid only in the area of stabs. If there is a strong acid (yellow colour) throughout the medium, a contaminating organism may be present. If in doubt about a tube containing a Neisseria species, a Gram stain and oxidase test should be performed on the growth.
- Storage / shelf life: The powder is very hygroscopic: store the powder at 10-30 °C, in a dry environment, in its original container tightly closed and use it before the expiry date on the label or until signs of deterioration or contamination are evident. Store prepared plates at 2-8 °C.
