Liofilchem CZAPEK DOX Agar Culture Media 610095 is a Liofilchem microbiology product supplied by Kormay Biomedicals. Its practical role is czapek Dox Agar is a medium used for the cultivation of fungi and bacteria capable of using inorganic nitrogen. Czapek Dox Agar is recommended by APHA for isolating Aspergillus, Penicillinum, Paecilomyces and related fungi, yielding characteristic mycelia and conidia. The only bacteria which are able to grow in this medium are non-fastidious soil bacteria.
This product is relevant to water and beverage laboratories where filtration, enrichment, colony recovery, and indicator-organism interpretation are tied to release or investigation decisions.

Principle behind the medium
Saccharose is the sole source of carbon and sodium nitrate is the sole nitrogen source. Dipotassium phosphate is the buffering agent. Magnesium sulfate, potassium chloride and ferrous sulfate provide essential ions. Agar is the solidifying agent.
How to build it into the SOP
- Preparation: Dehydrated medium Suspend 49 g of the powder in 1 liter of distilled or deionized water. Mix well. Heat to boil shaking frequently until completely dissolved. Sterilize in autoclave at 121°C for 15 minutes. Notice that the acidity of the medium may be increased to pH 3.5 ± 0.2 for the cultivation of acidophilic organisms, such as yeasts, by adding 10 ml of 10% lactic acid per liter after sterilization. Medium in bottles Melt the content of the bottle in a water bath at 100°C (loosing the cap partially removed) until completely dissolved.
- Procedure: Inoculate the medium by spreading the sample over the agar surface. Time and temperature of incubation vary considerably according to the species being cultivated. As a general guide, incubate for 1-2 weeks at 25°C. Most Penicillium spp have an optimum growth temperature between 20°C and 25°C, whilst many Aspergillus spp grow best at about 30°C. However, different fungi grow over a wide range of temperatures.

- Warnings / precautions: The product does not contain hazardous substances in concentrations exceeding the limits set by current legislation and therefore is not classified as dangerous. It is nevertheless recommended to consult the safety data sheet for its correct use. The product is intended for In vitro diagnostic use and must be used only by properly trained operators.
- Storage / shelf life: The powder is very hygroscopic, store the powder at 10-30°C, in a dry environment, in its original container tightly closed. Store bottles and prepared plates at 10-25°C away from light.
