Application snapshot
Liofilchem Listeria Aesculin Agar Culture Media 11230 is a Liofilchem microbiology product supplied by Kormay Biomedicals. Its practical role is lISTERIA AESCULIN AGAR is a selective medium used for the isolation and differentiation of Listeria spp in food and environmentalsamples, according to ISO 11290.
This product supports Listeria workflows in ready-to-eat food, dairy, meat, and environmental monitoring programs where early presumptive recovery must lead into confirmation.

Application fit for the lab
Use this product where the lab must protect a recovery step before confirmation. For ready-to-eat food and environmental samples, the important decision is not only whether growth appears, but whether the enrichment or plating step gives the organism enough opportunity to recover from stress, competing flora, preservatives, or cold-chain exposure.
Expected colonies or reaction pattern
Listeria spp appear concave, dark/black colored and surrounded by a halo of the same color.Suspect colonies must be submitted to Gram staining, biochemical tests, hemolysis tests on Columbia + 5% Sheep Blood, Camp test Sand Camp test R.
Principle behind the medium
Peptospecial, peptone, tryptose and maize starch supply nitrogen, carbon, amino acids and vitamins. Sodium chloride maintains theosmotic balance of the medium. Agar is the solidifying agent. The selectivity of the medium is due to cycloheximide, which inhibits thegrowth of fungi, and acriflavine, colistin, fosfomycin and cefotetan that inhibit bacterial flora. Esculin and ferric ammonium citrate allowthe presumptive identification of Listeria spp. that hydrolyze esculin to esculetin giving a dark color to the medium.
How to build it into the SOP
- Procedure: Streak a loopful of a suitable enrichment broth, incubated with the sample to be analyzed, on the surface of a plate. Invert the plate andincubate at 36+/-1° C for 24-48 hours.
