Method principle
Tryptone and soytone provide nitrogen and other nutrient to support microbial growth. Glucose is a source of carbohydrate. Yeast extract is a source of amino acids and vitamins of group B. Sodium chloride maintains the osmotic balance of the medium. Dipotassium phosphate is the buffer of the medium.
Bench workflow
- Preparation: Suspend 36.0 g of powder in 1 liter of distilled or deionized water. Heat until completely dissolved. Sterilize in autoclave at 121 °C for 15 minutes. Cool to 45-50°C. Aseptically add 2 vials of Listeria Supplement for Enrichment (ref. 81045).
- Procedure: Add 25 mL of liquid or 25 g of solid test material to 225 mL of LISTERIA ENRICHMENT BROTH and mix or blend thoroughly. Incubate at 30 ± 2°C for 48 hours. Using a sterile loop spread the incubated broth sample onto the entire surface of a selective agar plate for Listeria spp. Incubate at 36 ± 1 °C for 24-48 hours.
Storage, limits, and safety
- Warnings / precautions: The product is not classified as hazardous by current legislation and does not contain harmful substances in concentrations of ≥1%. The product must be used only by properly trained operators.
- Storage / shelf life: 10-30°C away from light, until the expiry date on the label or until signs of deterioration or contamination are evident. Store the prepared medium at 2-8 °C. WARNING and
