Liofilchem Listeria UVM 2 Broth Culture Media 610045 is a Liofilchem microbiology product supplied by Kormay Biomedicals. Its practical role is lISTERIA UVM media are prepared according to the formulations described by Donnelly and Baigent and by MacCalin and Lee. Listeria UVM 1 and 2 are used for the two steps enrichment of Listeria spp in meat products.

Bench workflow
- Procedure: Add 225 ml of Listeria UVM1 Broth to 25 g or 25 ml of sample. Homogenize for 2 minutes. Incubate at 30°C for 24 hours. Spread 0.2 ml of the incubated broth onto Listeria selective agar plates. After 24 hours add 1 ml to 4.5 ml of sterile KOH solution (2.5 g KOH and 20 g NaCl in 1000 ml of distilled water: pH over 12). Mix for 1 minute and then subculture onto a Listeria selective agar plates. After 24 hours transfer 0.1 ml of Listeria UVM1 culture to 10 ml of Listeria UVM2 Broth. Incubate the secondary enrichment medium at 30°C for 24 hours. Spread 0.2 ml of secondary enrichment medium on a Listeria selective agar plate. Add 1 ml to 4.5 ml of sterile KOH solution and proceed as before.

DIRECTIONS Suspend 54.4 g of powder in 1 liter of distilled or deionized water. Heat until completely dissolved. Dispense into final containers. Sterilize in autoclave at 115 °C for 15 minutes.
Storage, limits, and safety
- Storage / shelf life: The powder is very hygroscopic: store the powder at 10-30 °C, in a dry environment, in its original container tightly closed and use it before the expiry date on the label or until signs of deterioration or contamination are evident. Store prepared tubes at 2-8 °C.
