Principle behind the medium
Tryptone and yeast extract provide nitrogen and other nutrients necessary to support the bacterial growth. Sodium chloride keeps the osmotic balance of the medium. Sodium azide inhibits the bacterial flora grown during the preliminary test, except for enterococci. Glucose fermentation causes medium acidification with consequent medium color change from purple to yellow. The addition of glycerol increases glucose fermentation by enterococci.
How to build it into the SOP
- Preparation: Suspend 36.0 g of powder into 1 liter of distilled or deionized water. Add 5 mL of glycerol. Mix carefully. Dispense into final containers. Sterilize at 115 °C for 15 minutes.
- Procedure: For the performance of confirmation trial, transfer three loops of culture broth from each positive tube of AZIDE DEXTROSE BROTH into tubes of BROMOCRESOL PURPLE AZIDE BROTH. Incubate at 36+/-1 °C up to 48 hours.
