Bench workflow

  • Procedure: Collect the specimen to be tested in bottles containing sodium thiosulfate (about 100mg / l of specimen) in order to neutralise the possible residual chlorine. Inoculate five containers with 10 ml, five with 1 ml and five with 0.1 ml of the sample into Asparagine Enrichment Broth. Incubate at 36 ± 1 °C for 48 hours. The presence of P. aeruginosa is shown by turbidity and a blue-green fluorescent pigment, observed with a long-wave ultraviolet light.

Controls and performance checks

  • Formula / composition: (g/l) DL-Asparagine 3.0 Dipotassium Hydrogen Phosphate 1.0 Magnesium Sulphate 0.5 Final pH = 7.0 ± 0.2 at 25 °C. DIRECTIONS Suspend 4.5 g of powder in 1 liter of distilled or deionized water. Add 8 ml of glycerol, heat and shake until completely dissolved. Dispense into final containers. Sterilize in autoclave at 121 °C for 15 minutes.

  • Storage / shelf life: The powder is very hygroscopic: store the powder at 10-30 °C, in a dry environment, in its original container tightly closed and use it before the expiry date on the label or until signs of deterioration or contamination are evident. Store prepared media at 2-8 °C.
