Principle behind the medium
Peptones, dextrose and salts contained in the formulation, support the growth of microorganisms. Sodium azide inhibits gram-negative rods, whereas crystal violet is incorporated primarily to inhibit staphylococci.
How to build it into the SOP
- Preparation: Suspend 46 g in 1 litre of distilled or deionized water. Heat until completely dissolved. Sterilize in autoclave at 118°C for 15 minutes. Cool to 45-50°C and dispense in petri dishes.
- Procedure: Inoculate the sample by streaking onto the surface of the medium. Incubate for 24-48 hours at 36+/-1°C.
