Liofilchem BISMUTH SULPHITE Agar Culture Media 610301 is a Liofilchem microbiology product supplied by Kormay Biomedicals. Its practical role is bismuth Sulphite Agar is a medium used for the selective isolation of Salmonella spp, especially S. Typhi, from food, animal feedstuffs and environmental samples.

What to read after incubation

Salmonella Typhi appears as black colonies surrounded by a metallic sheen. Other Salmonella species have variable colonies which can be black, green, clear or mucoid. Other organisms, such as coliforms, Serratia and Proteus spp, are usually inhibited but occasionally there can be strains which give dull, green or brown colonies, with no metallic sheen.

Selective or enrichment logic

Meat extract, peptone and tryptone provide amino acids, nitrogen, carbon, vitamins and minerals for organisms growth. Glucose is the fermentable carbohydrate. Disodium phosphate is the buffering agent. Ferrous sulphate is an indicator of hydrogen sulfide production. Bismuth sulfite and brilliant green are selective agents effective against both Gram-positive and Gram-negative organisms other than Salmonella spp and some Shigella spp. Agar is the solidifying agent.

Sample handling workflow

  • Preparation: NOTE: Prepare plates on day before streaking and store in dark. Selectivity may decrease after 48h.
  • Procedure: Bismuth-Sulphite Agar may be used in conjunction with other selective agars for the isolation of salmonellae. Streak a loopful of the sample onto the surface of the agar. Incubate at 35 ± 2°C for 18-48 hours. Atypical colonies may appear if the medium is heavily inoculated with organic matter. To prevent such situation, suspend the sample in sterile saline and use the supernatant for inoculation. When the number of salmonellae is expected to be small, enrichments methods may be employed.

  • Storage / shelf life: The powder is very hygroscopic, store the powder at 10-30°C, in a dry environment, in its original container tightly closed. Store prepared plates at 2-8°C away from light. Do not use the product beyond its expiry date on the label or if product shows any evidence of contamination or any sign of deterioration.
  • Waste disposal: of waste must be carried out according to national and local regulations in force.
