Result signals and confirmation
Turbidity of the medium indicates microbial growth. To isolate colonies subculture to Salmonella Shigella Desoxycholate Calcium (SSDC) agar. After incubation at 30± 1°C for 18-24 hexamine the plates for suspect colonies. Final identification should be confirmed by standard biochemical tests.
Method principle
Enzymatic digest of casein provides amino acids, nitrogen, carbon, minerals, for organisms growth. Yeast extract is a source of vitamins, particularly of B-group. Sodium chloride maintains the osmotic balance of the medium. Irgasan (triclosan) inhibits Gram-positive bacteria and together with magnesium chloride and malachite green make the broth highly selective. Supplementation with Yersinia ITC Supplement (ref. 81092), containing ticarcillin and potassium chlorate, increases selectivity against both Gram-positive and Gram-negative bacteria.
Bench workflow
- Preparation: Suspend 44.1 g of powder in one liter of deionized or distilled water. Bring to boil and shake until completely dissolved. Sterilize at 121°C for 15 minutes. Cool up to 45-50°C. Aseptically, add rehydrated content of 2 vials (10 ml) of Yersinia ITC Supplement. Dispense into appropriate containers.
- Procedure: Prepare the initial suspension by adding the test sample to Yersinia ITC Broth to give a 1:100 dilution and homogenize for 2 min. Incubate at 25 ± 1°C for 40-48 h.
