Liofilchem Buffered Yeast Extract Broth Culture Media 610342 is a Liofilchem microbiology product supplied by Kormay Biomedicals. Its practical role is non-selective liquid medium used for the enrichment of Legionella species from environmental samples. This medium is not intended for use in the diagnosis of disease or other conditions in humans.

Application fit for the lab
This product is most useful when spoilage organisms or yeasts need controlled recovery and readable morphology. The lab should define incubation conditions, what counts as typical growth, and how suspect isolates are confirmed before the result is used for shelf-life or investigation decisions.
Expected colonies or reaction pattern
Turbidity indicates microbial growth in BYE Broth. For confirmation, subculture onto a plate of BCYE agar and a plate of BCYE without L-cysteine. Legionella species will grow on BCYE agar and will not grow on the same medium from which L-cysteine has been omitted. Growth on both plates indicates that the organism is not Legionella. Refer to the instructions for use of the agar media for more detailed information.
Principle behind the medium
Yeast extract provides amino acids, nitrogen, carbon, vitamins and minerals. Alpha-ketoglutarate stimulates organism growth. ACES buffer is included to maintain the proper pH for optimal growth. L-cysteine is an essential amino acid which is incorporated along with ferric pyrophosphate to satisfy specific nutritional requirements of Legionella species.
How to build it into the SOP
- Preparation: Suspend 21.65 g of the powder in 1 liter of distilled or deionized water. Mix well. Heat to boil shaking frequently until completely dissolved. Filter to sterilize. DO NOT AUTOCLAVE. Dispense into final containers, e.g. 10 ml tubes.
