Liofilchem Biggy (NICKERSON) Agar Culture Media 610135 is a Liofilchem microbiology product supplied by Kormay Biomedicals. Its practical role is selective and differential medium for the detection and isolation of Candida spp. from clinical samples. When this format is useful

This product is most useful when spoilage organisms or yeasts need controlled recovery and readable morphology. The lab should define incubation conditions, what counts as typical growth, and how suspect isolates are confirmed before the result is used for shelf-life or investigation decisions.

What to read after incubation

After incubation, observe the color and the morphology of the colonies for presumptive identification according to the following table. Microorganism Typical Colony Candida albicans Smooth, circular or hemispherical brown-black colonies; no color diffusion into surrounding medium; no metallic sheen. Candida tropicalis Smooth, discrete, dark brown to black colonies (may have black-colored centers); slight mycelial fringe; metallic sheen.

Selective or enrichment logic

Yeast extract provide amino acids, nitrogen, carbon, vitamins and minerals for organisms growth. Glycine stimulates growth. Glucose is the fermentable carbohydrate providing carbon and energy. Candida spp., through a process of substrate reduction, produce sulfide and bismuth which combine to produce brown to black pigmented colonies and zones of dark precipitate in the medium surrounding colonies of some species. Agar is the solidifying agent.

Sample handling workflow

  • Preparation: Dehydrated medium Suspend 47.0 g of the powder in 1 liter of distilled or deionized water. Heat to boil shaking frequently until completely dissolved. DO NOT AUTOCLAVE. Medium in bottles Melt the content of the bottle in a water bath at 100°C (losing the cap partially removed) until completely dissolved. Then screw the cap and check the homogeneity of the dissolved medium, if it is the case turning the bottle upside down. Cool at 45-50°C, mix well avoiding foam formation and aseptically distribute into Petri dishes.

Inoculate the plates by directly streaking the specimen on the agar surface or spread the sample from an enrichment culture to obtain well-isolated colonies.
